The p.Q167* pathogenic mutation (also known as c.499C>T), located in coding exon 4 of the TP53 gene, results from a C to T substitution at nucleotide position 499. This changes the amino acid from a glutamine to a stop codon within coding exon 4. This alteration was identified as a somatic alteration in AML and MDS tumors, and found to have between 0-10% of wildtype transactivation activity across multiple response elements tested (Lodé L et al. Haematologica, 2018 Jan;103:e13-e16). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
Li-Fraumeni syndrome Pathogenic:1
ClinVar contains an entry for this variant (Variation ID: 528250). This sequence change creates a premature translational stop signal (p.Gln167*) in the TP53 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in TP53 are known to be pathogenic (PMID: 20522432). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with gastric carcinoma or myelodysplastic syndromes (PMID: 21080251, 27741277). For these reasons, this variant has been classified as Pathogenic.
