This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with familial hypercholesterolemia, however in these individuals a pathogenic allele was also identified in LDLR (PMID: 9225977, 10737984, 14974088, 17142622, 25463123). This variant is also known as D365E. ClinVar contains an entry for this variant (Variation ID: 251693). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The glutamic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
