This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 496 of the LDLR protein (p.Gly496Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with familial hypercholesterolemia (PMID: 20809525; internal data). ClinVar contains an entry for this variant (Variation ID: 251867). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt LDLR protein function with a positive predictive value of 95%. This variant disrupts the p.Gly496 amino acid residue in LDLR. Other variant(s) that disrupt this residue have been observed in individuals with LDLR-related conditions (PMID: 26361156, 36648309), which suggests that this may be a clinically significant amino acid residue. For these reasons, this variant has been classified as Pathogenic.
