The encoded protein catalyzes the first step in hypusine formation by transferring the butylamine moiety of spermidine to a specific lysine residue of the eukaryotic translation initiation factor 5A precursor, forming an intermediate deoxyhypusine residue. The c.498C>G (p.I166M) alteration is located in exon 4 (coding exon 4) of the DHPS gene. This alteration results from a C to G substitution at nucleotide position 498, causing the isoleucine (I) at amino acid position 166 to be replaced by a methionine (M). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
