The p.Q305K variant (also known as c.913C>A), located in coding exon 1 of the CEBPA gene, results from a C to A substitution at nucleotide position 913. The glutamine at codon 305 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.

This variant is present in population databases (rs747606716, ExAC 0.01%). This sequence change replaces glutamine with lysine at codon 305 of the CEBPA protein (p.Gln305Lys). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and lysine. This variant has not been reported in the literature in individuals affected with CEBPA-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
