It has also been observed to segregate with disease in related individuals. This variant is present in population databases (rs200901862, gnomAD 0.01%). This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 7 of the TMEM147 protein (p.Gly7Arg). -

This alteration results from a G to C substitution at nucleotide position 19, causing the glycine (G) at amino acid position 7 to be replaced by an arginine (R). Based on data from gnomAD, the C allele has an overall frequency of 0.005% (14/279204) total alleles studied. The highest observed frequency was 0.014% (5/35396) of Latino alleles. This variant has been reported to be homozygous in three individuals from two families with features consistent with TMEM147-related neurodevelopmental disorder (Thomas, 2022). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
