Variant summary: RYR1 c.1534G>A (p.Glu512Lys) results in a conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251494 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1534G>A has been observed in individuals affected with clinical features of RYR1-related disorders (Wu_2006, Amburgey_2013, Invernizzi_2023, Jensson_2023). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 23919265, 37510298, 37937776, 16621918). ClinVar contains an entry for this variant (Variation ID: 65935). This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 512 of the RYR1 protein (p.Glu512Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of autosomal recessive RYR1-related conditions (PMID: 16621918).
