This variant has been observed in individual(s) with autosomal dominant central core disease (PMID: 11709545, Invitae). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 65962). This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with proline at codon 4793 of the RYR1 protein (p.Leu4793Pro). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline.
