Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Sep 04, 2024 | The p.G640R variant (also known as c.1918G>A), located in coding exon 13 of the CCDC114 gene, results from a G to A substitution at nucleotide position 1918. The glycine at codon 640 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. - |
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Dec 17, 2020 | In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0". The arginine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with CCDC114-related conditions. This variant is present in population databases (rs771734792, ExAC 0.01%). This sequence change replaces glycine with arginine at codon 640 of the CCDC114 protein (p.Gly640Arg). The glycine residue is weakly conserved and there is a moderate physicochemical difference between glycine and arginine. - |
