The c.32G>A (p.R11Q) alteration is located in exon 1 (coding exon 1) of the BBS5 gene. This alteration results from a G to A substitution at nucleotide position 32, causing the arginine (R) at amino acid position 11 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

The BBS5 c.32G>A variant is predicted to result in the amino acid substitution p.Arg11Gln. This variant was observed in an individual with ventricular tachycardia but did not segregate with disease (Golbus et al. 2014. PubMed ID: 25179549, supplementary table 7). We have observed this variant in the heterozygous state in an individual that had a homozygous BBS1 pathogenic variant (Internal Data). This variant is reported in 0.026% of alleles in individuals of European (Non-Finnish) descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
