The p.T603K variant (also known as c.1808C>A), located in coding exon 8 of the BARD1 gene, results from a C to A substitution at nucleotide position 1808. The threonine at codon 603 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. This variant is denoted BARD1 c.1808C>A at the cDNA level, p.Thr603Lys (T603K) at the protein level, and results in the change of a Threonine to a Lysine (ACA>AAA). This variant has not, to our knowledge, been published in the literature as a pathogenic or benign germline variant. BARD1 Thr603Lys was not observed at a significant allele frequency in large population cohorts (Lek 2016). This variant is located in the BRCT1 domain (Fox 2008, UniProt). In silico analysis, which includes protein predictors and evolutionary conservation, supports a deleterious effect. Based on currently available evidence, it is unclear whether BARD1 Thr603Lys is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. -
