The p.D458E variant (also known as c.1374C>A), located in coding exon 5 of the BARD1 gene, results from a C to A substitution at nucleotide position 1374. The aspartic acid at codon 458 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces aspartic acid, which is acidic and polar, with glutamic acid, which is acidic and polar, at codon 458 of the BARD1 protein (p.Asp458Glu).
