To our knowledge, functional studies have not been reported for this variant. In a large breast cancer case-control study, this variant has been observed in 3/60463 cases and 7/53454 controls; OR=0.379 (95%CI 0.098 to 1.465); p-value=0.206 (PMID: 33471991 - Leiden Open Variation Database DB-ID BARD1_000394). This variant has been identified in 2/263944 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.N215S variant (also known as c.644A>G), located in coding exon 4 of the BARD1 gene, results from an A to G substitution at nucleotide position 644. The asparagine at codon 215 is replaced by serine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -

Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was observed with an allele frequency of 3.2e-05 in 30954 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.644A>G in individuals affected with Hereditary Breast and Ovarian Cancer and no experimental evidence demonstrating its impact on protein function have been reported.
