The glycine at codon 759 is replaced by valine, an amino acid with dissimilar properties. While this exact alteration has not been reported in the literature, an alteration at the same codon, p.G759E (c.2276G>A), has shown <25% repair efficiency in an in vitro rapid cell-free assay (Drost M et al, Hum. Mutat. 2012 Mar; 33(3):488-94). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
