This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 759 of the MSH2 protein (p.Gly759Val). This variant has not been reported in the literature in individuals affected with MSH2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1005173). Advanced modeling of experimental studies (such as gene expression, population dynamics, functional pathways, and cell-cycle effects in cell culture) performed at Invitae indicates that this missense variant is expected to disrupt MSH2 protein function. Experimental studies have shown that this missense change affects MSH2 function (PMID: 33357406). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
Hereditary cancer-predisposing syndrome Uncertain:1
The p.G759V variant (also known as c.2276G>T), located in coding exon 14 of the MSH2 gene, results from a G to T substitution at nucleotide position 2276. The glycine at codon 759 is replaced by valine, an amino acid with dissimilar properties. While this exact alteration has not been reported in the literature, an alteration at the same codon, p.G759E (c.2276G>A), has shown <25% repair efficiency in an in vitro rapid cell-free assay (Drost M et al, Hum. Mutat. 2012 Mar; 33(3):488-94). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
