This changes the amino acid from a serine to a stop codon within coding exon 4. While this exact mutation has not been reported in the literature, a close match (c.2105C>A) giving rise to the same premature stop codon at amino acid position 702 has been reported in an individual diagnosed with colon, uterine, and ovarian cancer, with concordant tumor data showing loss of MSH6 via immunohistochemistry (Plaschke J et al. Hum. Mutat. 2004 Mar;23(3):285). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. - |
