The GNAS c.1462G>A variant is predicted to result in the amino acid substitution p.Ala488Thr. This variant is located in the pre-coding region of the primary GNAS transcript (NM_000516:c.-37000G>A). This variant has been reported in a healthy, ancestrally diverse cohort (Table S1, Bodian et al. 2014. PubMed ID: 24728327). This variant is reported in 0.022% of alleles in individuals of South Asian descent in gnomAD. Although we suspect this variant may be benign, at this time, the clinical significance is uncertain due to the absence of conclusive functional and genetic evidence. -
