This variant is present in population databases (rs140727644, gnomAD 0.04%). This missense change has been observed in individual(s) with primary ciliary dyskinesia (PMID: 24094744). ClinVar contains an entry for this variant (Variation ID: 525246). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). Experimental studies have shown that this missense change affects C21orf59 function (PMID: 24094744).
