This missense change has been observed in individuals with autosomal recessive autoimmune polyendocrinopathy syndrome (PMID: 17118990, 27253668). ClinVar contains an entry for this variant (Variation ID: 552873). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt AIRE protein function with a positive predictive value of 95%. This variant has been reported in multiple individuals with autosomal recessive autoimmune polyendocrine syndrome (Table 3, Wolff et al. 2007. PubMed ID: 17118990; Bruserud et al. 2016. PubMed ID: 27253668). This variant is reported in 0.0020% of alleles in individuals of European (non-Finnish) descent in gnomAD. Although we suspect that this variant may be pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
