Variant summary: DEPDC5 c.2576C>T (p.Thr859Met) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00011 in 249544 control chromosomes. To our knowledge, no occurrence of c.2576C>T in individuals affected with Epilepsy, Familial Focal, With Variable Foci 1 and no experimental evidence demonstrating its impact on protein function have been reported. Four submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance. -

This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 859 of the DEPDC5 protein (p.Thr859Met). This variant is present in population databases (rs200744555, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with DEPDC5-related conditions. ClinVar contains an entry for this variant (Variation ID: 534801). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
