MCM2 (HGNC:6944): (minichromosome maintenance complex component 2) The protein encoded by this gene is one of the highly conserved mini-chromosome maintenance proteins (MCM) that are involved in the initiation of eukaryotic genome replication. The hexameric protein complex formed by MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. This protein forms a complex with MCM4, 6, and 7, and has been shown to regulate the helicase activity of the complex. This protein is phosphorylated, and thus regulated by, protein kinases CDC2 and CDC7. Multiple alternatively spliced transcript variants have been found, but the full-length nature of some variants has not been defined. [provided by RefSeq, Oct 2012]

Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.0033 in 328844 control chromosomes (gnomAD and jMorp databases), predominantly at a frequency of 0.013 in a cohort of healthy Japanese individuals, including 9 homozygotes (jMorp datbase, ToMMo 38KJPN cohort; Tadaka_2021). The high allele frequency and presence of homozygotes in the Japanese subpopulation strongly suggest that the variant is benign. c.53G>C has been reported in the literature in individuals affected with polycystic ovary syndrome, however without strong evidence for causality (e.g., Zhou_2023). These reports do not provide unequivocal conclusions about association of the variant with Autosomal Dominant Nonsyndromic Hearing Loss 70. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 33179747, 37132453). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely benign. -
