MECOM (HGNC:3498): (MDS1 and EVI1 complex locus) The protein encoded by this gene is a transcriptional regulator and oncoprotein that may be involved in hematopoiesis, apoptosis, development, and cell differentiation and proliferation. The encoded protein can interact with CTBP1, SMAD3, CREBBP, KAT2B, MAPK8, and MAPK9. This gene can undergo translocation with the AML1 gene, resulting in overexpression of this gene and the onset of leukemia. Several transcript variants encoding a few different isoforms have been found for this gene. [provided by RefSeq, Mar 2011]

This sequence change has been described in the gnomAD database with a frequency of 0.0056% in the Latino/Admixed American subpopulation (dbSNP rs779316050). The p.Ala1040Val change affects a moderately conserved amino acid residue located in a domain of the MECOM protein that is not known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Ala1040Val substitution. This sequence change does not appear to have been previously described in individuals with MECOM-related disorders. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Ala1040Val change remains unknown at this time -
