OPA1 (HGNC:8140): (OPA1 mitochondrial dynamin like GTPase) The protein encoded by this gene is a nuclear-encoded mitochondrial protein with similarity to dynamin-related GTPases. The encoded protein localizes to the inner mitochondrial membrane and helps regulate mitochondrial stability and energy output. This protein also sequesters cytochrome c. Mutations in this gene have been associated with optic atrophy type 1, which is a dominantly inherited optic neuropathy resulting in progressive loss of visual acuity, leading in many cases to legal blindness. This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 396 of the OPA1 protein (p.Leu396Arg). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt OPA1 protein function. ClinVar contains an entry for this variant (Variation ID: 167403). This variant is also known as T1188G or c.1352T>G (p.Leu451Arg). This missense change has been observed in individuals with OPA1-related conditions (PMID: 12036970, 25205859, 29350691, 32855858). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency).
