This missense change has been observed in individuals with OPA1-related conditions (PMID: 12036970, 25205859, 29350691, 32855858). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 396 of the OPA1 protein (p.Leu396Arg). ClinVar contains an entry for this variant (Variation ID: 167403). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
