Genes affected
TGFBR2 (HGNC:11773): (transforming growth factor beta receptor 2) The protein encoded by this gene is a transmembrane protein that has a protein kinase domain, forms a heterodimeric complex with TGF-beta receptor type-1, and binds TGF-beta. This receptor/ligand complex phosphorylates proteins, which then enter the nucleus and regulate the transcription of genes related to cell proliferation, cell cycle arrest, wound healing, immunosuppression, and tumorigenesis. Mutations in this gene have been associated with Marfan Syndrome, Loeys-Deitz Aortic Aneurysm Syndrome, and the development of various types of tumors. Alternatively spliced transcript variants encoding different isoforms have been characterized. [provided by RefSeq, Aug 2017]

Uncertain significance, criteria provided, single submitterclinical testingGeneDxJan 17, 2018A variant of uncertain significance has been identified in the TGFBR2 gene. The c.-27 G>C variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016). The c.-27 G>C variant is located in the 5' untranslated region of the TGFBR2 gene at a nucleotide that is conserved through mammals. Other variants in the 5' regulatory region of the TGFBR2 gene have been reported in HGMD in association with TGFBR2-related disorders (Stenson et al., 2014). However, this variant does not create a new start codon and does not alter the Kozak sequence. Nevertheless, the in absence of functional studies the consequence of the c.-27 G>C variant cannot be precisely determined. -
Diabetic retinopathy Other:1
Uncertain risk allele, criteria provided, single submitterresearchClinical Genomics, Uppaluri K&H Personalized Medicine Clinic-Potent mutations in TGFBR2 gene encodes the transforming growth factor that have been associated with angiogenesis and diabetic retinopathy. More clinical studies are needed for stronger association. However, more evidence is required to confer the association of this particular variant rs1469986980 with diabetic retinopathy. -
