TGFBR2 (HGNC:11773): (transforming growth factor beta receptor 2) The protein encoded by this gene is a transmembrane protein that has a protein kinase domain, forms a heterodimeric complex with TGF-beta receptor type-1, and binds TGF-beta. This receptor/ligand complex phosphorylates proteins, which then enter the nucleus and regulate the transcription of genes related to cell proliferation, cell cycle arrest, wound healing, immunosuppression, and tumorigenesis. Mutations in this gene have been associated with Marfan Syndrome, Loeys-Deitz Aortic Aneurysm Syndrome, and the development of various types of tumors. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has been observed in individuals with clinical features of Loeys-Dietz syndrome, Marfan syndrome, or thoracic aortic aneurysm and dissection (PMID: 24793577, 27879313, Invitae). ClinVar contains an entry for this variant (Variation ID: 165392). This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with proline at codon 339 of the TGFBR2 protein (p.Arg339Pro). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and proline. -
