The glycine at codon 351 is replaced by aspartic acid, an amino acid with similar properties, and is located in the protein kinase domain. This variant has been reported in individuals diagnosed with Loeys-Dietz syndrome (Maleszewski JJ et al. Am. J. Surg. Pathol., 2009 Feb;33:194-201; Wang WJ et al. J. Mol. Med., 2013 Jan;91:37-47; Frischmeyer-Guerrerio PA et al. Sci Transl Med, 2013 Jul;5:195ra94). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic. - |
