TGFBR2 (HGNC:11773): (transforming growth factor beta receptor 2) The protein encoded by this gene is a transmembrane protein that has a protein kinase domain, forms a heterodimeric complex with TGF-beta receptor type-1, and binds TGF-beta. This receptor/ligand complex phosphorylates proteins, which then enter the nucleus and regulate the transcription of genes related to cell proliferation, cell cycle arrest, wound healing, immunosuppression, and tumorigenesis. Mutations in this gene have been associated with Marfan Syndrome, Loeys-Deitz Aortic Aneurysm Syndrome, and the development of various types of tumors. Pathogenic, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Feb 08, 2017 | Family studies have indicated that this variant was not present in the parents of an individual with symptoms consistent with TGFBR2-related disease, which suggests that it was de novo in that affected individual (Invitae). For these reasons, this variant has been classified as Pathogenic. A different missense substitution at this codon (p.Ser449Phe) is reported to be deleterious (PMID: 21098638) and has been reported in individuals with Marfan syndrome and Loeys-Dietz syndrome (PMID: 15235604, 18781618,22113417, 23884466, 16928994). This indicates that the serine residue is important for TGFBR2 protein function. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies. This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with cysteine at codon 449 of the TGFBR2 protein (p.Ser449Cys). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and cysteine. - |
