The encoded protein is a small GTPase that contains both N-terminal and C-terminal guanine nucleotide-binding motifs. This protein is localized in the cilia and plays a role in cilia formation and in maintenance of cilia. Mutations in this gene are the cause of Joubert syndrome 8. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.461A>G has been reported in the literature in at least one compound heterozygous individual affected with Joubert Syndrome (e.g.Bachmann-Gagescu_2015). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two submitters have provided clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as pathogenic (n=1)/likely pathogenic (n=1).
