Variant summary: ANK2 c.2472_2474dupCAC (p.Thr826dup) results in an in-frame duplication that is predicted to duplicate one amino acid into the encoded protein. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 249986 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2472_2474dupCAC in individuals affected with Long QT Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance. -

The c.2472_2474dupCAC variant (also known as p.T826dup), located in coding exon 22 of the ANK2 gene, results from an in-frame duplication of CAC at nucleotide positions 2472 to 2474. This results in the duplication of an extra residue between codons 826 and 827. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
