The variant allele was found at a frequency of 4e-06 in 251338 control chromosomes (gnomAD). c.1366C>T has been reported in the literature in multiple individuals affected with Glutaric Aciduria, Type 2c (example: Behin_2016 and Lupica_2022). Other variant(s) that disrupt this residue is associated with Glutaric Aciduria, Type 2c in HGMD. These data indicate that the variant is very likely to be associated with disease. Four clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and all classified the variant as likely pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

This variant is present in population databases (rs751821289, gnomAD 0.002%). This missense change has been observed in individuals with multiple acyl-CoA dehydrogenase deficiency (PMID: 27038534, 35309592; Invitae). ClinVar contains an entry for this variant (Variation ID: 374577). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ETFDH protein function with a positive predictive value of 80%. This variant disrupts the p.Pro456 amino acid residue in ETFDH. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 17412732, 17584774, 23727839, 26403312). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. -
