The protein functions in aneural activation of muscle-specific receptor kinase, which is required for postsynaptic differentiation, and in the subsequent clustering of the acetylcholine receptor in myotubes. This protein can also induce autophosphorylation of muscle-specific receptor kinase. Mutations in this gene are a cause of familial limb-girdle myasthenia autosomal recessive, which is also known as congenital myasthenic syndrome type 1B. The variant allele was found at a frequency of 8e-06 in 250644 control chromosomes. c.514G>A has been reported in the literature in compound heterozygous individuals affected with Congenital Myasthenic Syndrome and in one family, this variant has been shown to segregate with disease (Ben Ammar_2010, Gaist_2017, Normand_2018, Ek_2023). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 20012313, 37273706, 28716243, 30266093). ClinVar contains an entry for this variant (Variation ID: 560992).
