TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. This receptor functionally interacts with toll-like receptor 2 to mediate cellular response to bacterial lipoproteins. The c.1994A>C (p.E665A) alteration is located in exon 2 (coding exon 1) of the TLR6 gene. This alteration results from a A to C substitution at nucleotide position 1994, causing the glutamic acid (E) at amino acid position 665 to be replaced by an alanine (A). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
