This variant is present in population databases (rs145602440, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with KIT-related conditions. The p.R804W variant (also known as c.2410C>T), located in coding exon 17 of the KIT gene, results from a C to T substitution at nucleotide position 2410. The arginine at codon 804 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
