Genes affected
SNCA (HGNC:11138): (synuclein alpha) Alpha-synuclein is a member of the synuclein family, which also includes beta- and gamma-synuclein. Synucleins are abundantly expressed in the brain and alpha- and beta-synuclein inhibit phospholipase D2 selectively. SNCA may serve to integrate presynaptic signaling and membrane trafficking. Defects in SNCA have been implicated in the pathogenesis of Parkinson disease. SNCA peptides are a major component of amyloid plaques in the brains of patients with Alzheimer's disease. Alternatively spliced transcripts encoding different isoforms have been identified for this gene. [provided by RefSeq, Feb 2016]

ACMG classification

Classification was made for transcript

Our verdict: Pathogenic. The variant received 10 ACMG points.

PS3
PS3 evidence extracted from ClinVar submissions: SCV002572882: Functional studies provide strong evidence of the variant having a damaging effect on the gene or gene product (PMID: 24728187, 24984882).
PM1
In a hotspot region, there are 5 aminoacids with missense pathogenic changes in the window of +-8 aminoacids around while only 0 benign, 2 uncertain in NM_000345.4
PM2
Very rare variant in population databases, with high coverage;
PP5
Variant 4-89828154-C-T is Pathogenic according to our data. Variant chr4-89828154-C-T is described in ClinVar as Pathogenic. ClinVar VariationId is 97000.Status of the report is criteria_provided_single_submitter, 1 stars.
