Variant summary: TERT c.2455C>A (p.Arg819Ser) results in a non-conservative amino acid change located in the Reverse transcriptase domain (IPR000477) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 248072 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2455C>A in individuals affected with Dyskeratosis congenita and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 1413839). Based on the evidence outlined above, the variant was classified as uncertain significance. -

This variant is present in population databases (rs746621306, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with TERT-related conditions.
