The p.R788H variant (also known as c.2363G>A), located in coding exon 18 of the FBN2 gene, results from a G to A substitution at nucleotide position 2363. The arginine at codon 788 is replaced by histidine, an amino acid with highly similar properties. This alteration has been reported in a hereditary aortopathy cohort (Renner S et al. Genet Med, 2019 08;21:1832-1841). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
