Therefore, it has been classified as a Variant of Uncertain Significance. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals affected with SLC22A4-related conditions. This variant is present in population databases (rs773811534, gnomAD 0.007%). This sequence change replaces arginine, which is basic and polar, with threonine, which is neutral and polar, at codon 166 of the SLC22A4 protein (p.Arg166Thr). This variant also falls at the last nucleotide of exon 2, which is part of the consensus splice site for this exon. - |
