In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SLC22A5 protein function. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This sequence change replaces arginine, which is basic and polar, with proline, which is neutral and non-polar, at codon 282 of the SLC22A5 protein (p.Arg282Pro). This variant is present in population databases (no rsID available, gnomAD 0.004%). For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Arg282 amino acid residue in SLC22A5. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 16652335, 20574985, 21922592).
