Genes affected
GEMIN5 (HGNC:20043): (gem nuclear organelle associated protein 5) This gene encodes a WD repeat protein that is a component of the survival of motor neurons (SMN) complex. The SMN complex plays a critical role in mRNA splicing through the assembly of spliceosomal small nuclear ribonucleoproteins (snRNPs), and may also mediate the assembly and transport of other classes of ribonucleoproteins. The encoded protein is the snRNA-binding component of the SMN complex. Dysregulation of this gene may play a role in alternative mRNA splicing and tumor cell motility. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Nov 2011]

The c.4263-1G>C variant is not present in 1000 Genomes, EVS, ExAC, gnomAD, Indian Exome Database or our in-house exome database. This variant has neither been published in the literature in individuals affected with GEMIN5-related conditions nor reported to the ClinVar, HGMD or OMIM databases. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In-silico pathogenicity prediction programs like HSF3.1, MutationTaster2, CADD, Varsome, Franklin, etc. predicted this variant to be likely deleterious; however, these predictions were not confirmed by published translational/functional studies. This variant was identified in an individual as a part of extended carrier screening. -
