ClinVar contains an entry for this variant (Variation ID: 159331). For these reasons, this variant has been classified as Pathogenic. -
This sequence variant is a single nucleotide substitution (C>T) at position 4417 of the NSD1 gene that results in the generation of an early termition codon at residue 1473 of the NSD1 protein. This variant is predicted to result in a non-functiol allele through either the expression of a truncated protein or a loss of NSD1 expression due to nonsense mediated decay. This is a previously reported variant (ClinVar) that has been observed in multiple individiuals affected with Sotos Syndrome (PMID: 15942875).
