This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 13 of the MAK protein (p.Gly13Ser). This variant is present in population databases (rs387906647, gnomAD 0.002%). This missense change has been observed in individual(s) with retinitis pigmentosa (PMID: 21835304, 25385675). ClinVar contains an entry for this variant (Variation ID: 29783). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt MAK protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects MAK function (PMID: 21835304).
