The encoded protein is a component of the SWI/SNF chromatin remodeling complex and may play a role in cell-cycle activation. The protein encoded by this locus is similar to AT-rich interactive domain-containing protein 1A. These two proteins function as alternative, mutually exclusive ARID-subunits of the SWI/SNF complex. The p.R2128* pathogenic mutation (also known as c.6382C>T), located in coding exon 20 of the ARID1B gene, results from a C to T substitution at nucleotide position 6382. This changes the amino acid from an arginine to a stop codon within coding exon 20. This variant was identified de novo in an individual with Coffin-Siris syndrome (Wieczorek D et al. Hum. Mol. Genet., 2013 Dec;22:5121-35). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation. As such, this alteration is interpreted as a disease-causing mutation. -
