This protein stimulates ERCC3/XPB ATPase activity to trigger DNA opening during DNA repair, and is implicated in regulating cellular levels of TFIIH. Mutations in this gene result in trichothiodystrophy, complementation group A. [provided by RefSeq, Mar 2009]

Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with GTF2H5-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with valine at codon 20 of the GTF2H5 protein (p.Leu20Val). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and valine. -
