Mutations in this gene are known to cause Parkinson disease and autosomal recessive juvenile Parkinson disease. Alternative splicing of this gene produces multiple transcript variants encoding distinct isoforms. Additional splice variants of this gene have been described but currently lack transcript support. The p.Ala230Val variant has allele frequency 0.02% in gnomAD. This variant has been submitted to the ClinVar database as Uncertain Significance. The amino acid change p.Ala230Val in PRKN is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Ala at position 230 is changed to a Val changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as Variant of Uncertain Significance (VUS). In the absence of another reportable variant in PRKN gene, the molecular diagnosis is not confirmed. - |

The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and valine. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies. In summary, this variant is a novel missense change that is not predicted to affect protein function. There is no indication that it causes disease, but the available evidence is currently insufficient to prove that conclusively. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is present in population databases (rs571490973, ExAC 0.2%) but has not been reported in the literature in individuals with a PARK2-related disease. - |
