The ENST00000950058.1(SERPINE1):c.-123+152_-123+153insC variant causes a intron change. Note: ENST00000950058.1 is not a MANE Select or MANE Plus Clinical transcript for SERPINE1; the reported annotation may differ from that of the MANE-designated reference transcript for this gene. The gene SERPINE1 is a tumor suppressor gene (CancerMine: 2 TSG, 10 oncogene, 2 driver citations). The gene SERPINE1 is a known oncogene (CancerMine: 2 TSG, 10 oncogene, 2 driver citations). The gene SERPINE1 is a cancer driver gene (CancerMine: 2 TSG, 10 oncogene, 2 driver citations). The variant allele was found at a cumulative frequency of 0.0000658 (AC=10) in the gnomAD database across 151,922 control chromosomes (no homozygotes observed). The grpmax filtering allele frequency (95% CI) is 0.0000117. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar.

Genes affected
SERPINE1 (HGNC:8583): (serpin family E member 1) This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. The protein also functions as a component of innate antiviral immunity. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia.
