Variant summary: SLC26A4 c.1343C>T (p.Ser448Leu) results in a non-conservative amino acid change located in the SLC26A/SulP transporter domain (IPR011547) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 2e-05 in 251068 control chromosomes. c.1343C>T has been reported in the literature in multiple individuals affected with Pendred Syndrome (examples: (Kahrizi_2015, Ideura_2019, Liu_2020).). These data indicate that the variant is very likely to be associated with disease. The following publications have been ascertained in the context of this evaluation (PMID: 18813951, 31427586, 32447495). ClinVar contains an entry for this variant (Variation ID: 549979). Based on the evidence outlined above, the variant was classified as pathogenic. -
