This sequence change replaces proline, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 59 of the GCK protein (p.Pro59Ala). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of maturity onset diabetes of the young (internal data). ClinVar contains an entry for this variant (Variation ID: 1341600). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt GCK protein function with a negative predictive value of 80%. This variant disrupts the p.Pro59 amino acid residue in GCK. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 22060211, 22335469).
