Mutations in this gene have been associated with complementation group 1 peroxisomal disorders such as neonatal adrenoleukodystrophy, infantile Refsum disease, and Zellweger syndrome. The variant was absent in 247966 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.3833dupA in individuals affected with Zellweger Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. Other variants causing extension of the protein or frameshift at the C-terminal are cited as uncertain significance in ClinVar (e.g. p.Ser1270LysfsX12, p.Asn1268LysfsX23). Nevertheless, a variant that alters the normal termination codon leading to extension of the protein (p.X1284GlnextX28) has been reported in the literature in an individual affected with neonatal adrenoleukodystrophy (PMID: 11389485). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014.
