Multiple congenital exostosis Pathogenic:1
This premature translational stop signal has been observed in individual(s) with multiple exostoses (PMID: 29620724; Invitae). For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. ClinVar contains an entry for this variant (Variation ID: 191230). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Trp711*) in the EXT1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 36 amino acid(s) of the EXT1 protein. -
